目的:研究XW630对人的类成骨细胞株TE85细胞功能的影响。方法:[~3H]胸腺嘧啶掺入法和细胞计数法测定细胞增殖、放射免疫法测定cGMP含量、精氨酸转化法测定iNOS活性。结果:在培养的人的类成骨细胞中,XW630作用48h后,细胞数增加96.9%,[~3H]胸腺嘧啶掺入量增加62.7%。NOS抑制剂L-NMMA(0.25,0.5,1.0 mmol·L~(-1))作用48h,可剂量依赖地抑制细胞增殖,这一作用可被XW630 1.0nmol·L~(-1)部分阻断。作用时间在30min内,XW630对细胞iNOS活性及cGMP含量无影响。延长作用时间,XW630时间依赖地增加细胞iNOS活性及cGMP含量。结论:XW630刺激人的类成骨细胞增殖,增加细胞iNOS活性和cGMP含量。
Objective: To study the effect of XW630 on the function of human osteoblast cell line TE85. Methods: [~ 3H] thymidine incorporation and cell counting were used to determine the cell proliferation. Radioimmunoassay was used to determine the cGMP content. The activity of iNOS was determined by arginine conversion assay. Results: In cultured human osteoblasts, XW630 increased the number of cells by 96.9% and increased the incorporation of [~ 3H] thymine by 62.7% after 48 hours. NOS inhibitor L-NMMA (0.25,0.5,1.0 mmol·L -1) for 48 h inhibited cell proliferation in a dose-dependent manner, which was blocked by 1.0 nmol·L -1 XW630 . The action time within 30min, XW630 had no effect on iNOS activity and cGMP content. Prolonging the action time, XW630 increased cell iNOS activity and cGMP content in a time-dependent manner. Conclusion: XW630 stimulates human osteoblast proliferation and increases iNOS activity and cGMP content.
