目的:研究次乌头碱与甘草苷合用对心肌兴奋-收缩偶联相关离子通道基因mRNA表达的影响。方法:成年Wistar大鼠分别灌胃次乌头碱(0.23、0.69、2.07mg.kg-1.d-1),甘草苷(20mg.kg-1.d-1),以及次乌头碱(2.07mg.kg-1.d-1)与甘草苷(20mg.kg-1.d-1)7d后,应用实时荧光定量PCR检测各给药组心肌组织中Cav1.2、NCX、SCN5A和RyR2mRNA的表达水平。结果:次乌头碱灌胃给药后能显著抑制NCX mRNA的表达与显著诱导RyR2mRNA的表达;甘草苷仅对NCX mRNA的表达有显著抑制作用;甘草苷与次乌头碱合用不仅能显著抑制SCN5A mRNA的表达与显著诱导RyR2mRNA的表达,也能显著拮抗高剂量次乌头碱对NCX mRNA表达的抑制作用。结论:甘草苷可通过干预次乌头碱对大鼠心肌SCN5A、NCX和RyR2mRNA表达的影响,从而影响次乌头碱导致的心律失常。
Objective: To study the effect of hypaconitine and glycyrrhizin on the mRNA expression of ion channel related to excitatory-contractile coupling in myocardium. Methods: Adult Wistar rats were administered intragastric hypaconitine (0.23,0.69,2.07mg.kg-1.d-1), glycyrrhizin (20mg.kg-1.d-1) and hypaconitine Cav1.2, NCX, SCN5A and RyR2mRNA in myocardial tissue of each drug-treated group were detected by real-time fluorescence quantitative PCR after 7 days, 2.07mg.kg-1.d-1 and glycyrrhizin (20mg.kg-1.d- The level of expression. Results: Hypaconitine could inhibit the expression of NCX mRNA and RyR2 mRNA significantly after intragastric administration. Glycyrrhizin only inhibited the expression of NCX mRNA significantly. Administration of glycyrrhizin and hypaconitine could not only significantly inhibit the expression of NCX mRNA, The expression of SCN5A mRNA and the significant expression of RyR2 mRNA also significantly inhibited the inhibitory effect of high dose hypaconitine on NCX mRNA expression. Conclusion: Glycyrrhizin can affect the hypaconitine-induced arrhythmia through the intervention of hypaconitine on the expression of SCN5A, NCX and RyR2 mRNA in the myocardium of rats.
