目的 :探讨糖尿病妊娠时胎鼠胰 β细胞中分泌颗粒 ( β颗粒 )的变化。方法 :取 Wistar大鼠制作糖尿病妊娠模型 ,于妊娠第 17、18、19、2 0天取胎鼠胰腺 ,进行透射电镜观察及立体重建等立体定量分析。结果 :正常对照组第 19天 β颗粒的所有参数显著大于第 18天的 ;而在实验组 ,18天 β颗粒的所有参数显著大于第 17天的。实验组于对照组比较 ,第 17、18天 β颗粒的平均直径、平均体积、平均表面积及体密度较对照组的显著增加 ,而且其 18天 β颗粒的数面密度、数密度也较对照组的显著增加。结论 :妊娠糖尿病可促进胎鼠胰 β细胞颗粒提前成熟。
Objective: To investigate the changes of secretory granules (β granules) in fetal rat pancreatic β cells in diabetic pregnancy. Methods: Wistar rats were used to make gestational diabetes mellitus. Pancreas of fetus was taken on the 17th, 18th, 19th and 20th day of gestation, and then subjected to three-dimensional quantitative analysis by transmission electron microscopy and three-dimensional reconstruction. Results: All parameters of β-granule on day 19 of the normal control group were significantly greater than those on the 18th day. In the experimental group, all parameters of the beta-particle on day 18 were significantly greater than on the 17th day. Compared with the control group, the average diameter, average volume, average surface area and body density of the β-granule in the experimental group were significantly increased on the 17th and 18th days, and the number-average density and the number density of the β-granule on the 18th day were also higher than those in the control group A significant increase. Conclusion: Gestational diabetes mellitus can promote premature maturation of fetal rat pancreatic β-cell particles.
