[目的]该研究旨在筛选一套适于辣椒杂交种纯度鉴定的核心SSR引物。[方法]利用17对SSR引物对100份已知辣椒杂交种进行DNA指纹分析。[结果]根据多态性和杂合率两个指标,确定Hpms1-214、Es395和Hpms1-5为辣椒杂交种纯度鉴定的首选核心引物。利用这3个引物进行筛选,97个品种(占97%)能够找到具有杂合带型的鉴定引物,确定5个引物Es330、Es363、Epms923、Es120和Es64为辣椒杂交种纯度鉴定的备选核心引物。筛选出14个品种的特异性引物,可进一步筛选每个辣椒杂交种的双亲互补型引物。[结论]该研究为构建辣椒杂交种纯度鉴定标准DNA指纹图谱奠定基础。
[Objective] The aim of this study was to screen a set of core SSR primers suitable for purity identification of pepper hybrids. [Method] DNA fingerprinting of 100 capsicum hybrids was conducted using 17 pairs of SSR primers. [Result] According to the polymorphism and heterozygosity, Hpms1-214, Es395 and Hpms1-5 were identified as the preferred core primers for purity identification of pepper hybrids. Using these three primers for screening, 97 primers (97%) were able to find the identification primers with heterozygous bands. The five primers Es330, Es363, Epms923, Es120 and Es64 were identified as the alternative cores for purity identification of pepper hybrids Primer. Fourteen varieties of specific primers were screened to further screen for the parents of each pepper hybrid. [Conclusion] The research laid the foundation for constructing the DNA fingerprinting of purity identification standard of pepper hybrids.
