目的:观察冠心合剂对兔颈动脉经皮冠状动脉腔内成形术(PTCA)损伤后细胞外基质Ⅰ型胶原蛋白表达的影响。方法:运用球囊原位扩张、拉伤致兔颈总动脉损伤建立PTCA术后内皮损伤模型。从术前3天开始各治疗组予相应剂量冠心合剂灌胃给药,分别于2、4、6周处死动物,免疫组织化学法检测Ⅰ型胶原的表达。结果:各中药治疗组内膜胶原密度较模型组降低,差异有统计学意义(P<0.05);冠心合剂大剂量组6周的内膜胶原密度表达最少。结论:冠心合剂可保护兔颈动脉球囊损伤后内膜,其机制可能与减少血管壁胶原含量有关。
Objective: To observe the effect of Guanxin Mixture on the expression of extracellular matrix type Ⅰ collagen (Collagen) after carotid artery percutaneous transluminal coronary angioplasty (PTCA) in rabbits. Methods: Endothelial injury model of PTCA was established by balloon dilatation and injury of carotid artery in rabbits. From the 3 days before surgery, each treatment group was given the corresponding dose of Guanxin Mixture by intragastric administration, and the animals were sacrificed at 2, 4 and 6 weeks respectively. The expression of type Ⅰ collagen was detected by immunohistochemistry. Results: The density of the intima collagens in each TCM group was lower than that in the model group (P <0.05). At the 6th week of Guanxin Mixture high dose group, the density of the intima collagens was the least. Conclusion: Guanxin Mixture can protect the intima after carotid artery balloon injury in rabbits, and its mechanism may be related to the reduction of collagen content in the blood vessel wall.
