目的:探讨赖氨酰氧化酶在大鼠急性脊髓损伤中的修复作用。方法健康雌性Wistar大鼠30只建立急性脊髓损伤中平分为实验组、对照组、空白组,实验组损伤前大鼠饮用水中给予氯化钴(30mg/L),对照组进行造模后注射同剂量的生理盐水,空白组仅进行开关腹操作,不对脊髓及血管进行操作。结果实验组脊髓损伤后6h和24h的Tarlov评分明显高于对照组(<0.05);同时两组的Tarlov评分在各个时间点的评分都明显高于空白组(<0.05)。在脊髓损伤48h后实验组与对照组的赖氨酰氧化酶表达量都明显高于空白组(<0.05),同时对照组的赖氨酰氧化酶表达量明显低于实验组(<0.05)。结论赖氨酰氧化酶在大鼠急性脊髓损伤中呈现高表达状况,提高赖氨酰氧化酶的表达能促进脊髓损伤的修复。“,”Objective To investigate the ef ect of lysyl oxidase (LOX) in repair process of acute spinal cord injury (SCI) in rats. Methods 96 healthy female SD rats were divided into four groups:Group A, damaged rats treated with β-aminopropionitrile (BAPN, an inhibitor of LOX);Group B, damaged rats treated with cobalt chloride (CoCl2 , an enhancer of LOX);Group C, damaged rats with no any treatment (only injury);Group D, normal rats (sham),Using the Al en 's methods to establish the acute spinal cord injury model for rats, and set ing up the same hit ing force. Injured spinal cords were removed for protein extraction at dif erent time after injure (1,3,7, 14, and 30 days).The expression of LOX in tissues with spinal cord injury was detected by western blot, and analyzing the significance between group A,B,C and D. Correlating with the recovery of hind limb movement evaluated by the BBB methods. Results The results of BBB score show that the grade of group B was obviously higher than that of group A and C. The of Western blot show that compared with group D, the expression of lox increased in group B of 1 d, peaked at 3d, decreased at 7d, and decreased significantly at 14d and 30d; Compared with group B, the expression curve of LOX in group A is basical y the same, but the expression rate was significantly lower than the lat er. In group C the expression of LOX peaked at 1d, and reached the minimum at 7 d, and recovered at 14 d and 30 d. The expression of LOX in group B was obviously higher than group A and C, and it is statistical y significant between the two group at the same time ( <0.05). Conclusion The upregulation of LOX expression and bet er recovery of hind limb movement in SCI rats treated with CoCl2 suggest that LOX may play an important role in repair process of spinal cord injury.
