目的:探讨DLL4、Notch1在结肠癌中的表达及其临床意义。方法:收集十堰市人民医院病理科2009年1月至2012年10月存档的结肠蜡块标本95例,其中正常黏膜组织8例,结肠癌70例,包括高分化腺癌30例、中分化腺癌25例、低分化腺癌15例;结肠腺瘤17例。运用免疫组织化学SP法检测DLL4、Notch1在组织中的表达,分析其表达水平与临床病理之间的关系。结果:(1)在结肠正常黏膜一腺瘤-癌序列中,DLL4的阳性率呈递增趋势,其在结肠癌组中的表达率高于正常黏膜组(P<0.05);(2)Notch-1的阳性率在结肠正常黏膜-腺瘤-癌序列中亦呈递增趋势。其在结肠癌组中的表达率高于正常黏膜组(P<0.05);(3)DLL4、Notch-1在结肠癌不同分化程度中的表述率不同,分化越低,阳性率越高(P<0.01)。结论:DLLA/Notch1的表达与结肠癌的发生、发展有关,可为结肠癌基因治疗提供靶点。
Objective: To investigate the expression of DLL4 and Notch1 in colon cancer and its clinical significance. Methods: A total of 95 specimens of colon wax were archived from January 2009 to October 2012 in Department of Pathology, People’s Hospital of Shiyan, including 8 cases of normal mucosa and 70 cases of colon cancer, including 30 cases of well-differentiated adenocarcinoma, 25 cases of cancer, poorly differentiated adenocarcinoma in 15 cases; colon adenoma in 17 cases. The expression of DLL4 and Notch1 in tissues was detected by immunohistochemical SP method. The relationship between the expression of DLL4 and clinicopathological features was analyzed. Results: (1) The positive rate of DLL4 in colorectal mucosa-adenocarcinoma-cancer sequence showed an increasing trend, which was higher in colon cancer group than in normal mucosa group (P <0.05). (2) Notch- 1 positive rate in the normal colon mucosa - adenoma - cancer sequence also showed an increasing trend. (3) The expressions of DLL4 and Notch-1 in different differentiation degree of colon cancer were different, the lower the differentiation was, the higher the positive rate was (P <0.05) <0.01). Conclusion: The expression of DLLA / Notch1 is related to the occurrence and development of colon cancer and may provide a target for the gene therapy of colon cancer.
