为了解中国地区宫颈癌病人中人乳头瘤病毒16 型E6E7 基因结构特点,从中国山东地区宫颈癌活检组织中提取组织DNA,经HPV 多重引物PCR 法鉴定标本中感染HPV 型别,选单纯感染HPV16 型两例标本DNA为模板进行PCR 扩增,获得HPV16 E6E7 基因后,重组入pALTER-1 载体,进行双向测序、分析。DNA 序列分析表明:两例标本的HPV16 E6E7 序列全长均为776bp, 与已发表的德国标准株长度相等,两例均为变异株,共检出5 处核苷酸变异位点,其中4 处可导致氨基酸改变。中国山东地区宫颈癌病人中HPV16 E6E7 的基因结构与德国标准株的HPV16 E6E7 基因之间存在差异。
In order to understand the structural characteristics of human papillomavirus type 16 E6E7 gene in cervical cancer patients in China, tissue DNA was extracted from cervical cancer biopsy tissue in Shandong Province of China. HPV types were identified by PCR with multiplex PCR of HPV, and HPV16 Type two specimens of DNA as a template for PCR amplification, obtained HPV16 E6E7 gene, recombinant pALTER-1 vector for bidirectional sequencing and analysis. DNA sequence analysis showed that the HPV16 E6E7 sequences of both samples were all 776bp in length, which was equal to the length of the published German standard strain, and two of them were mutants. Five nucleotide variation sites were detected, of which 4 Can lead to amino acid changes. The genetic structure of HPV16 E6E7 in patients with cervical cancer in Shandong Province of China differs from the HPV16 E6E7 gene in German standard strains.
