目的:为了探讨血小板活化因子(PAF)及其受体对颈髓损伤后颈髓血管内皮细胞细胞间粘附分子1mRNA(ICAM1mRNA)和内皮细胞白细胞粘附分子1mRNA(ELAM1mRNA)表达的影响。方法:通过Alen’s法造成鼠C67挫伤,采用鞘内注射PAF及静脉注射PAF受体拮抗剂BN52021,应用地高辛标记cRNA探针原位杂交技术检测颈髓损伤后颈髓血管内皮细胞上述粘附分子的表达。结果:伤后6~12小时颈髓血管内皮细胞ICAM1mRNA、ELAM1mRNA表达呈不同程度的增加;鞘内注射PAF可使伤后血管内皮细胞ICAM1mRNA、ELAM1mRNA表达明显增加,PAF受体拮抗剂可抑制ICAM1mRNA、ELAM1mRNA表达。结论:PAF通过增加伤后颈髓血管内皮细胞粘附分子的表达是导致血脊髓屏障损害的重要分子基础。PAF受体对伤后血管内皮细胞细胞粘附分子的表达具有调控作用。
Objective: To investigate the effect of platelet activating factor (PAF) and its receptor on the expression of intercellular adhesion molecule1 mRNA (ICAM1 mRNA) and endothelial cell leukocyte adhesion molecule1 mRNA (ELAM1 mRNA) in cervical spinal cord endothelial cells The impact of expression. Methods: C6.7 contusion was induced by Alen’s method. PAF was injected intrathecally and PA5 receptor antagonist BN52021 was injected intravenously. Digoxin-labeled cRNA probe in situ hybridization was used to detect the changes of cervical intravascular endothelial Cell expression of the above adhesion molecules. Results: ICAM 1mRNA and ELAM-1mRNA expression in cervical vascular endothelial cells were increased in 6 ~ 12h after injury; intrathecal injection of PAF could significantly increase the expression of ICAM1mRNA and ELAM1mRNA in injured vascular endothelial cells, PAF Receptor antagonist can inhibit I CAM 1 mRNA, ELAM 1 mRNA expression. CONCLUSION: PAF is an important molecular basis for the damage of blood-spinal cord barrier by increasing the expression of adhesion molecules in injured cervical spinal cord endothelial cells. PAF receptor plays a regulatory role in the expression of cell adhesion molecules in injured vascular endothelial cells.
