目的:探讨超级化激活环核苷酸调控通道基因家族-2(HCN2)基因在风湿性心脏病二尖瓣狭窄合并心房颤动患者右心耳组织中的表达。方法:采用实时荧光定量聚合酶链反应(FQ-PCR)的方法分析HCN2mRNA在43例风湿性二尖瓣狭窄患者右心耳组织中的表达,并对反应产物进行测序。结果:测定HCN2基因cDNA序列同源性为100%,风湿性心脏病二尖瓣狭窄合并心房颤动组HCN2mRNA表达量明显高于窦性心律组(P<0.01),HCN2mRNA的表达与左心房内径、心胸比例呈正相关,而与二尖瓣瓣口面积呈负相关。结论:风湿性心脏病二尖瓣狭窄患者并发心房颤动的发生发展过程中,HCN2mRNA表达明显上调。
Objective: To investigate the expression of HCN2 gene in the right atrial appendage of patients with rheumatic mitral stenosis and atrial fibrillation. Methods: The expression of HCN2 mRNA in right atrial appendage of 43 rheumatic mitral stenosis patients was analyzed by real-time fluorescence quantitative polymerase chain reaction (FQ-PCR). The reaction products were sequenced. Results: The homology of HCN2 cDNA sequence was 100%. The expression of HCN2 mRNA in rheumatic mitral stenosis with atrial fibrillation was significantly higher than that in sinus rhythm group (P <0.01). The expression of HCN2 mRNA was correlated with the diameter of left atrium, Cardiothoracic proportion was positively correlated with mitral valve orifice area was negatively correlated. Conclusion: The expression of HCN2 mRNA in patients with rheumatic mitral stenosis complicated with atrial fibrillation was significantly up-regulated.
