目的研究乳源性多肽对新生儿抗感染作用,为生命早期健康成长提供营养保障。方法利用质谱定量技术比较β-Casein-15肽在早产和足月产产妇母乳中的含量差异;通过Uniprot、SABLE、ProtParam tool在线数据库预测β-Casein-15肽生物学特征;以抑菌圈实验以及免疫荧光实验来检测β-Casein-15肽对大肠杆菌、小肠结肠炎菌、金黄色葡萄球菌的抑菌作用。结果通过质谱分析以及生物信息学分析发现:β-Casein-15肽的脂肪指数是122.33,亲水性是-0.387,不稳定系数是36.29。这些数据表明β-Casein-15肽是稳定性疏水性多肽。质谱定量分析发现β-Casein-15肽在早产儿产妇母乳中含量明显高于足月儿产妇,抑菌实验发现β-Casein-15肽对大肠杆菌、小肠结肠炎菌以及金黄色葡萄球菌生长均有明显的抑制作用。结论这些结果提示β-Casein-15肽在抗新生儿感染中发挥着重要的作用。
Objective To study the anti-infective effect of milk-derived peptides on newborn and to provide nutrition support for the healthy growth of early life. Methods The mass fraction of β-Casein-15 peptide in preterm and full-term mothers was compared by mass spectrometry. The biological characteristics of β-Casein-15 peptide were predicted by on-line database of Uniprot, SABLE and ProtParam tool. And immunofluorescence assay to detect the inhibitory effect of β-Casein-15 peptide on Escherichia coli, enterocolitis and Staphylococcus aureus. Results The results of mass spectrometry and bioinformatics analysis showed that the β-Casein-15 peptide had a fat index of 122.33, a hydrophilicity of -0.387 and an unstable coefficient of 36.29. These data indicate that the β-Casein-15 peptide is a stable hydrophobic polypeptide. Mass spectrometry quantitative analysis found β-Casein-15 peptide content in preterm children was significantly higher than that of full-term mothers, antibacterial experiments found β-Casein-15 peptide on Escherichia coli, enterocolitis bacteria and Staphylococcus aureus growth were Obvious inhibitory effect. Conclusions These results suggest that β-Casein-15 peptide plays an important role in anti-neonatal infection.
