采用脱氧核苷酸转移酶介导的缺口末端标记法,观察兔腹主动脉经球囊导管损伤后0、8、24h,3天和5天时血管中膜平行肌细胞和外膜细胞凋亡的变化.提取血管组织RNA,采用反转录一多聚酶链反应测定一氧化氮合成酶基因表达。结果表明,血管损伤后8h可见中膜平滑肌细胞凋亡增加,24h达高峰,3天和5天仍维持在较高水平,外膜细胞凋亡无明显改变。而诱导型一氧化氮合成酶基因表达亦于8h明显增加,在24h达最高,与细胞凋亡存在平行关系。提示中膜平滑肌细胞凋亡是球囊导管损伤血管的一个早期反应,诱导型一氧化氮合成酸可能参与了此过程。
The apoptosis of the intima-media and intima-media of the tunica intima was observed at 0, 8, 24, 3, and 5 days after balloon catheter injury in the abdominal aorta of rabbits by using deoxynucleotidyl transferase mediated nick end labeling Variety. RNA was extracted from vascular tissue and nitric oxide synthase gene expression was determined by reverse transcription-polymerase chain reaction. The results showed that 8 h after vascular injury, the apoptosis of VSMC increased, reached the peak at 24 h, remained high at 3 and 5 days, and there was no obvious change in the apoptosis of adventitia. The expression of inducible nitric oxide synthase gene was also significantly increased at 8h, reached the highest at 24h, and there was a parallel relationship between apoptosis. It is suggested that apoptosis of the smooth muscle cells of the tunica media is an early response to vascular injury of balloon catheters. It is suggested that inducible nitric oxide synthase may be involved in this process.
