目的利用蛋白质组学技术识别结晶型硫化镍(NiS)诱发人支气管上皮细胞恶性转化相关蛋白的差异表达,探讨镍致癌的分子机制。方法应用二维凝胶电泳与相应软件分析结晶型NiS诱发细胞恶性转化后蛋白质组变化,基质辅助激光解吸电离-飞行时间-质谱(MALDITOFMS)结合数据库检索鉴定恶性转化相关蛋白。结果获得分辨率和重复性较好的二维凝胶电泳图谱,在相对分子质量14400~94000、等电点3~10范围内分离出约700个不同蛋白质点,对差异表达明显的等电点约6,相对分子质量约25000的蛋白质点进行质谱分析,鉴定出等电点为5.66,相对分子质量为21890的蛋白硫氧还原蛋白过氧化物酶2(Peroxiredoxin2,PDX2),并发现该蛋白在恶性转化细胞中高表达。结论蛋白PDX2可能是参与结晶型NiS诱发人支气管上皮细胞恶性转化过程的重要蛋白分子,与细胞恶性转化有关。
OBJECTIVE: To identify the differential expression of malignant transformation-associated proteins in human bronchial epithelial cells induced by crystalline nickel sulfide (NiS) by proteomics and to explore the molecular mechanism of nickel carcinogenesis. Methods Two-dimensional gel electrophoresis and corresponding software were used to analyze proteomic changes of malignant transformed cells induced by crystalline NiS. Malignant transformation related proteins were identified by MALDI-TOF-MS. Results Two-dimensional gel electrophoresis with good resolution and repeatability was obtained. Approximately 700 different protein spots were isolated in the range of molecular weight 14400-94000 and isoelectric point 3-10, and the isoelectric points About 6, the relative molecular mass of about 25000 protein spots were analyzed by mass spectrometry to identify the isoelectric point of 5.66, the relative molecular mass of 21890 protein thioredoxin peroxidase 2 (Peroxiredoxin2, PDX2) and found that the protein in Highly expressed in malignant transformed cells. Conclusion Protein PDX2 may be an important protein involved in the malignant transformation of human bronchial epithelial cells induced by crystalline NiS, which is related to the malignant transformation of cells.
