目的:考察益阴通络胶囊对局灶性脑缺血模型大鼠血清丙二醛(MDA);乳酸脱氢酶(LDH)及脑组织血管内皮生长因子(VEGF)的影响。方法:采用大脑中动脉阻塞法制备大鼠局灶性脑缺血模型,将造模成功后大鼠按神经学评分随机分为假手术组、模型对照组、阳性对照组、益阴通络胶囊大、中、小剂量组六组,给药两周后进行神经功能学评分、测定血清SOD和MDA含量和免疫组织化学法测定脑组织中VEGF。结果:益阴通络胶囊大剂量组(10.8g/kg)和中剂量组(5.4g/kg)可降低局灶性脑缺血模型大鼠神经行为学评分(P<0.01);大剂量组(10.8g/kg)、中剂量组(5.4 g/kg)和小剂量组(2.7g/kg)可降低血清MDA和LDH含量(P<0.05,P<0.01);大剂量组(10.8g/kg)、中剂量组(5.4g/kg)和小剂量组(2.7g/kg)可增加脑组织中VEGF含量(P<0.05)。结论:益阴通络胶囊对大鼠局灶性脑缺血有保护作用,其作用机制可能与抗自由基、促进新生血管形成有关。
Objective: To investigate the effects of Yiyin Tongluo Capsule on serum malondialdehyde (MDA), lactate dehydrogenase (LDH) and vascular endothelial growth factor (VEGF) in rats with focal cerebral ischemia. Methods: The model of focal cerebral ischemia in rats was established by middle cerebral artery occlusion (MCAO). The rats were randomly divided into sham operation group, model control group, positive control group, Yiyin Tongluo Capsule The rats in large, medium and small dose groups were scored for neurological function two weeks after the administration, the levels of SOD and MDA in serum were measured, and the levels of VEGF in brain tissue were measured by immunohistochemistry. Results: Yiyintongluo capsule high dose group (10.8g / kg) and middle dose group (5.4g / kg) could reduce the neurobehavioral score of focal cerebral ischemia model rats (P <0.01); high dose group (10.8g / kg), medium dose (5.4g / kg) and low dose (2.7g / kg) decreased MDA and LDH levels in serum (P <0.05, kg), middle dose (5.4g / kg) and low dose (2.7g / kg) increased the content of VEGF in brain tissue (P <0.05). Conclusion: Yiyin Tongluo Capsule has a protective effect on focal cerebral ischemia in rats. Its mechanism may be related to anti-free radical and angiogenesis.
