用5只正常犬进行了国产头孢三嗪的药代动力学和组织浓度研究。剂量均为20mg/kg,浓度以微生物法测定。 结果显示,肌肉和静脉注射的药代动力学分别符合开放型一室和二室模型。肌注后0.75小时血浓度达峰值(58.76μg/ml),生物利用度完全。静脉注射后峰值为171.11μg/ml。肌注和静注的生物半衰期分别为2.63和2.74小时;表观分布容积分别为0.4和0.422L/kg。 4只犬作了组织浓度测定。结果显示胆汁浓度最高,达1933.68μg/ml,肾脏浓度次之。其它组织如子宫、肝、前列腺、肺、小肠、心、脾及肌肉的浓度依次减低,但均超过多数敏感菌株的最低杀菌浓度。脑组织浓度为0.18μg/g。
Five normal dogs were used to study the pharmacokinetics and tissue concentration of domestic ceftriaxone. The dosage was 20mg / kg, the concentration of microbiological determination. The results showed that the pharmacokinetics of intramuscular and intravenous injections were in line with the open-studio and two-compartment models, respectively. 0.75 hours after intramuscular injection of blood peak (58.76μg / ml), bioavailability completely. The peak after intravenous injection was 171.11 μg / ml. The biological half-lives for intramuscular and intravenous injection were 2.63 and 2.74 hours, respectively; the apparent volume of distribution was 0.4 and 0.422 L / kg, respectively. Four dogs were tested for tissue concentration. The results showed that the highest bile concentration, up to 1933.68μg / ml, followed by the kidney concentration. Other tissues, such as uterus, liver, prostate, lung, small intestine, heart, spleen and muscle concentration in order, but more than the most sensitive strains of the minimum bactericidal concentration. Brain tissue concentration was 0.18 μg / g.
