目的观察41℃60min温热作用后人胃癌细胞BGC-823热耐受性和HSP70表达的变化。方法41℃60min温热作用后不同时间,测定细胞43℃60min温热再作用的克隆形成率,并进行细胞HSP70免疫组化分析。结果①41℃诱导后即时细胞已形成较高的热耐受性,4h有所降低,8h达到高峰(P<0.01),24h恢复近原有水平;②BGC-823细胞常温下表达少量HSP70,并且主要分布于胞浆内;41℃诱导后即时HSP70的表达量并未增加,但核内染色加深(P<0.01);诱导后4h开始增加(P<0.01),8~16h达到高峰,并主要在核内(P<0.01);24h衰减至近原有水平(P>0.05)。结论41℃温热诱导后BGC-823细胞形成的热耐受性包括两部分:一部分为即时、非HSP70依赖的,另一部分为HSP70依赖的热耐受性。
Objective To observe the changes of heat tolerance and HSP70 expression in human gastric cancer cells BGC-823 after warming at 41°C for 60 min. Methods The rate of colony formation in cells at 43°C for 60 min after warming was measured at 41°C for 60 min. The cloning rate was measured and the HSP70 immunohistochemical analysis was performed. RESULTS: Immediately after 141°C induction, the cells had a high thermal tolerance, which decreased at 4 h, peaked at 8 h (P<0.01), and recovered to the original levels at 24 h; 2 BGC-823 cells expressed a small amount of HSP70 at room temperature. And mainly distributed in the cytoplasm; immediately after induction at 41 °C, the expression of HSP70 did not increase, but the nuclear staining increased (P<0.01); it increased at 4 hours after induction (P<0.01), 8 to 16 hours. Peaks were reached and mainly in the nucleus (P<0.01); 24h decayed to near original levels (P>0.05). Conclusion The heat tolerance of BGC-823 cells induced by warming at 41°C includes two parts: one is immediate, non-HSP70 dependent, and the other is HSP70-dependent thermal tolerance.
