抑癌基因p53增强胶质瘤自杀基因作用的实验研究

目的 研究抑癌基因p53提高恶性胶质瘤自杀基因治疗的可行性. 方法 以C6鼠胶质瘤细胞和SD大鼠建立动物模型,原位注射腺病毒为载体的p53和自杀基因,腹腔给以阿昔洛韦(100 mg·kg-1day-1 × 14 d);观察荷瘤鼠生存期:强化MRI动态监测荷瘤鼠肿瘤大小;TUNEL法检测凋亡;评价P53基因对自杀基因系统治疗胶质瘤的优化作用. 结果 体内p53基因联合自杀基因使荷瘤鼠生存期明显延长,凋亡明显增加;强化MRI示肿瘤生长明显减慢. 结论 在体内p53基因可明显增强胶质瘤自杀基因系统的抑瘤作用.
