本试验以盆栽3年生新红星/海棠为试材,研究了苹果新根细胞分裂素的提取、纯化和高效形相色谱(HPLC)分析方法测定。苹果新根用80%冷甲醇抽提,在pH3.0下用石油醚、乙酸乙酯和在PHS.0~8.5下用正丁醇萃取,提取滚浓缩后用PH3.0的磷酸缓冲液做溶剂,加PVP振荡吸附,最后用SeP—PakC18简柱进行纯化,以甲醇、水溶剂系统(PH3.5)在SpherisorbC18柱上进行分离,用紫外检测器在265mm波长下检测、鉴定.苹果新根中的CTK种类主要有Z、ZR、diHZ。
In this study, potted three-year-old New Star / Begonia as test material, the apple new root cytokinin extraction, purification and high performance liquid chromatography (HPLC) analysis method. Fresh roots of apple are extracted with 80% cold methanol, petroleum ether, ethyl acetate and pH at pH 3.0. 0 ~ 8.5 with n-butanol extraction, extraction and rolling concentrated with PH3.0 phosphate buffer solvent, add PVP oscillation adsorption, and finally with SeP-PakC18 Jane column purification, methanol, water solvent system (PH3 .5) Separation on a Spherisorb C18 column, detection with UV detector at 265 mm wavelength, identification. CTK species in the new apple root are mainly Z, ZR, diHZ.
