目的研究不同剂量托吡酯(TPM)干预下,大鼠海马组织中神经细胞黏附分子(NCAM)和生长相关蛋白-43(GAP-43)mRNA的表达变化。方法将48只大鼠随机分成正常对照组、海人酸(KA)组、10mg/kgTPM组、40mg/kgTPM组、100mg/kgTPM组和400mg/kgTPM组,每组8只;建立不同剂量的TPM干预癫癎大鼠模型。观察大鼠行为学改变;通过Real-time PCR测定各组大鼠海马组织中NCAM和GAP-43的mRNA表达。结果KA组大鼠海马NCAM及GAP-43的mRNA表达水平明显高于正常对照组(P<0.01),10mg/kg TPM组和40mg/kg TPM组与KA组的差异无统计学意义,100mg/kg TPM组和400mg/kgTPM组明显低于KA组(P<0.01),其中400mg/kg TPM组明显低于100mg/kgTPM组(P<0.01)。结论KA诱导癫癎大鼠海马NCAM和GAP-43 mRNA表达上调,较大剂量TPM能够抑制NCAM和GAP-43 mRNA的表达,且抑制作用与TPM剂量有关。
Objective To investigate the expression changes of neural cell adhesion molecule (NCAM) and growth associated protein-43 (GAP-43) mRNA in hippocampus of rats with different doses of topiramate (TPM). Methods 48 rats were randomly divided into normal control group, KA group, 10mg / kg TPM group, 40mg / kg TPM group, 100mg / kg TPM group and 400mg / kg TPM group, with 8 rats in each group. Different doses of TPM Intervention in epileptic rat model. The behavioral changes of rats were observed. The mRNA expression of NCAM and GAP-43 in hippocampus were detected by Real-time PCR. Results The mRNA expression levels of NCAM and GAP-43 in hippocampus of rats in KA group were significantly higher than those in normal control group (P <0.01). There was no significant difference between KA group and 10mg / kg TPM group and 40mg / kg TPM group. kg TPM group and 400mg / kg TPM group was significantly lower than that of KA group (P <0.01), 400mg / kg TPM group was significantly lower than 100mg / kg TPM group (P <0.01). Conclusions KA induced elevation of NCAM and GAP-43 mRNA in hippocampus of rats with epilepsy, and higher doses of TPM can inhibit the expression of NCAM and GAP-43 mRNA.
