用胰岛细胞作同种同基因异体移植,是使Streptozotocin诱发的糖尿病大鼠得到好转的一种满意办法。使糖尿病获得缓解所需的胰岛细胞移植数量以及最佳的注输途径,已经明确。然而,对于用胶原酶-Ficoll溶液方法分离胰岛细胞的得率低,仍无明显进展。本文分析决定胰岛细胞最佳得率的各种因素。资料和方法:用200~250g的成年Lewis大鼠作为供体和受体。每一只受体用6只供体提供的胰腺制成移植材料。将受体动物分成11组,每组10只作比较。第1~3组比较胰腺碎块的大小,分别用1、2和>2mm的胰腺碎块。第4~6组比较制备胰岛细胞的pH值,分别用pH6.0、7.4、8.4的Hanks’溶液。第7、8组比
Islet allotrootransplantation with islet cells is a satisfactory solution to the improvement of Streptozotocin-induced diabetic rats. The number of islet cell transplants needed for the relief of diabetes and the optimal route of infusion have been established. However, little progress has been made in the isolation of pancreatic islet cells using collagenase-Ficoll solution. This article analyzes the factors that determine the optimal yield of islet cells. Materials and Methods: Adult Lewis rats weighing 200-250 g were used as donors and recipients. Pancreatic graft material was provided from 6 donors per recipient.
