目的:研究当归补血汤对动脉血管内皮细胞凋亡的影响,探讨其可能作用机制。方法:当归补血汤提取液作用于血管紧张素Ⅱ(AngⅡ)诱导的大鼠动脉内皮细胞(RAOEC)凋亡模型(细胞密度1×105/mL)24 h,检测RAOEC凋亡率和半胱氨酸蛋白酶-8(caspase-8),Bcl-2相关x蛋白(Bax),B细胞淋巴瘤/白血病-2(Bcl-2)mRNA的表达。结果:浓度为500 mg.L-1的当归补血汤作用于AngⅡ诱导的RAOEC凋亡细胞后,凋亡率为(8.69±0.69)%,与对照组(14.45±1.87)%相比,凋亡率明显下降,且差异有统计学意义(P<0.05);当归补血汤组RAOECs的caspase-8,Bax mRNA水平,Bax mRNA/Bcl-2 mRNA分别为0.603±0.081,0.629±0.079,0.566±0.113,均明显低于对照组(1.083±0.115,1.024±0.072,1.018±0.111),且差异具有统计学意义(均为P<0.05)。结论:当归补血汤具有抗动脉血管内皮细胞凋亡的作用;当归补血汤可能通过下调caspase-8和Bax基因表达分别从死亡受体通路和线粒体通路抑制RAOEC凋亡。
Objective: To study the effect of Dangguibuxuetang on the apoptosis of arterial endothelial cells and to explore its possible mechanism. Methods: The effect of Dangguibuxuetang extract on the apoptosis of rat aortic endothelial cells (RAOEC) induced by Ang Ⅱ (cell density 1 × 105 / mL) for 24 h, the apoptosis rate of RAOEC and cysteamine Caspase-8, Bcl-2 and Bcl-2 mRNA. Results: The apoptosis rate of Dangguibuxuetang with concentration of 500 mg · L-1 was (8.69 ± 0.69)% when compared with that of control group (14.45 ± 1.87)% (P <0.05). The expressions of caspase-8 and Bax mRNA and Bax mRNA / Bcl-2 mRNA in RAOECs of Dangguibuxue Decoction group were 0.603 ± 0.081, 0.629 ± 0.079 and 0.566 ± 0.113 (1.083 ± 0.115, 1.024 ± 0.072 and 1.018 ± 0.111 respectively), and the difference was statistically significant (both P <0.05). Conclusion: Dangguibuxuetang can prevent apoptosis of aortic endothelial cells. Dangguibuxuetang decoction can inhibit the apoptosis of RAOEC by downregulating the expression of caspase-8 and Bax from death receptor pathway and mitochondrial pathway, respectively.
