目的:探讨细丝蛋白A(FLNa)在肝癌患者外周血和癌组织中的表达及其与临床病理特征之间的关系。方法:将156例患者分为肝癌组和正常对照组,其中肝癌组97例,正常对照组59例(包括肝血管瘤患者47例,肝内胆管结石7例,肝挫裂伤5例);收集两组患者的血清及组织标本,用酶联免疫吸附(ELISA)法检测两组患者外周血中FLNa的含量,免疫组化法检测组织标本中FLNa的表达情况。结果:肝癌组患者血清中FLNa的平均浓度为(31.64±10.51)ρg/mL,正常对照组为(47.16±18.62)ρg/mL,肝癌组患者血清中FLNa水平明显低于正常对照组,P<0.05;FLNa在肝癌组织中同样低表达,肝癌组中FLNa的阳性表达率为21.64%,低于正常对照组的83.05%,两组相比差异有统计学意义,P<0.05。AFP值较高、分化程度较低、肿瘤直径较大以及发生复发转移的肝癌患者,FLNa在血清中的含量及组织中的表达显著降低,P<0.05;而不同性别、不同年龄段的肝患者血清中及组织中FLNa的表达水平,差异无统计学意义,P>0.05。结论:FLNa在肝癌患者外周血及癌组织中的表达显著降低,FLNa的低表达可能与肝癌的发生发展密切相关。
Objective: To investigate the expression of FLNa in peripheral blood and cancer tissues of patients with hepatocellular carcinoma and its relationship with clinicopathological features. Methods: A total of 156 patients were divided into HCC group and normal control group. 97 cases of HCC group and 59 cases of normal control group included 47 cases of hepatic hemangioma, 7 cases of intrahepatic bile duct stones and 5 cases of liver contusion and laceration. Serum and tissue samples from both groups were collected. The levels of FLNa in peripheral blood were detected by enzyme-linked immunosorbent assay (ELISA), and the expression of FLNa was detected by immunohistochemistry. Results: The average level of FLNa in serum of patients with hepatocellular carcinoma was (31.64 ± 10.51) ρg / mL and that of normal controls was (47.16 ± 18.62) ρg / mL, while the level of FLNa in patients with hepatocellular carcinoma was significantly lower than that of normal controls (P < 0.05). The positive rate of FLNa expression in HCC was 21.64%, which was lower than that in normal control group (83.05%). There was significant difference between the two groups (P <0.05). The levels of FLNa in serum and the expression of FLNa were significantly decreased in patients with hepatocellular carcinoma with higher AFP value, lower degree of differentiation, larger tumor diameter and recurrence and metastasis (P <0.05). However, patients with liver cancer of different gender and age There was no significant difference in the expression of FLNa in serum and tissues (P> 0.05). CONCLUSION: The expression of FLNa in peripheral blood and cancerous tissues of patients with HCC is significantly decreased. The low expression of FLNa may be closely related to the occurrence and development of HCC.
