This article describes a new method for the identification of conformational epitopes of Japanese encephalitis virus (JEV) .Fmoc chemistry was used to synthesize a complete pentapeptide library containing 20 to 5 pentapeptide sequences. The McAbs N.03 and N.08 were screened by immunoaffinity.It was found that the type sequences of the peptides with affinity to N.03 and N.08 were (Y / W / F) GG (I / L / M) and (N / Q) WY (D / E). In order to select the high affinity peptide, the pseudopeptide library (PPL) was further synthesized and for N.03 XXGGOXX (X, representing any of the 20 natural amino acids ; O, representing the specific amino acid for each reaction.)
