目的分析垂体瘤转化基因(PTTG)和结肠癌转移相关因子1(MACC1)在视网膜母细胞瘤(retinoblastoma,RB)中的表达与RB临床病理特征的关系。方法采用免疫组化方法检测PTTG和MACC1在42例RB肿瘤组织和正常视网膜组织中的表达,分析二者在不同临床特征和高危病理特征RB中的表达特征与关系。结果 PTTG在RB组织中表达丰富,但在正常视网膜中几乎不表达,二者差异有统计意义(χ~2=27.68,P<0.05);MACC1在RB组织和正常视网膜中都有表达,二者在表达程度上差异有统计意义(χ~2=9.62,P<0.05);PTTG在未分化型和分化型、脉络膜受侵和未受侵、视神经受侵和未受侵RB组织中的表达差异均有统计意义(χ~2=6.17,χ~2=6.99,χ~2=9.70,P<0.05);MACC1在未分化型和分化型、脉络膜受侵和未受侵RB组织中的表达差异无统计意义(χ~2=0.48,χ~2=2.10,P>0.05),但在视神经受侵和未受侵RB组织中的表达差异有统计意义(χ~2=8.52,P<0.05);PTTG和MACC1在不同性别、年龄之间的表达差异无统计意义(P>0.05);PTTG和MACC1在RB组织内的表达无显著相关性。结论 PTTG和MACC1在RB组织中高表达,而且表达水平与RB分化程度、扩散转移等高危病理特征密切相关,提示PTTG和MACC1在RB的发生、发展中具有一定作用,对于预测RB浸润和转移有重要价值。
Objective To analyze the relationship between the expression of pituitary tumor transforming gene (PTTG) and colon cancer metastasis related factor 1 (MACC1) in retinoblastoma (RB) and clinicopathological features of RB. Methods Immunohistochemistry was used to detect the expression of PTTG and MACC1 in 42 cases of RB tumor and normal retinal tissue. The expression characteristics and relationship of PTTG and MACC1 in RB with different clinical features and high risk pathological features were analyzed. Results PTTG was abundantly expressed in RB tissues but scarcely expressed in normal retina. The difference was statistically significant (χ ~ 2 = 27.68, P <0.05). The expression of MACC1 in both RB tissues and normal retina, both The difference was statistically significant (χ ~ 2 = 9.62, P <0.05). The difference of PTTG expression between undifferentiated and differentiated, choroidal invaded and noninvasive, optic nerve and noninvasive RB tissues (Χ ~ 2 = 6.17, χ ~ 2 = 6.99, χ ~ 2 = 9.70, P <0.05). The difference of MACC1 expression in undifferentiated and differentiated, choroidal invaded and noninvasive RB tissues (Χ ~ 2 = 0.48, χ ~ 2 = 2.10, P> 0.05), but there was a significant difference in the expression between optic nerve and noninvasive RB tissues (χ ~ 2 = 8.52, P <0.05) There was no significant difference in the expression of PTTG and MACC1 between different sexes and ages (P> 0.05). There was no significant correlation between the expressions of PTTG and MACC1 in RB tissues. Conclusions PTTG and MACC1 are highly expressed in RB tissues, and their expression levels are closely related to the high-risk pathological features such as RB differentiation and diffusion metastasis, suggesting that PTTG and MACC1 play a role in the development and progression of RB and are important for predicting RB infiltration and metastasis value.
