目的:观察羌活地黄汤含药血清对软骨细胞增值及RANKL mRNA(receptor activator nuclear factor kappa B ligand)表达的影响。方法:采用血清药理学和体外培养兔关节软骨细胞的方法,通过MTS/PMS系统检测羌活地黄汤含药血清对软骨细胞增殖的影响以及采用实时荧光定量PCR检测软骨细胞RANKL mRNA表达变化。结果:羌活地黄汤含药血清对体外培养的兔关节软骨细胞有促进增殖的作用,与实验对照组相比差异均有统计学意义(P<0.05),羌活地黄汤含药血清组RANKL mRNA表达量低于同一时间节点的实验对照组。结论:羌活地黄汤含药血清能促进体外培养的软骨细胞增殖,同时能抑制软骨细胞RANKL mRNA的表达。
OBJECTIVE: To observe the effect of the serum containing Zhan Hudi Decoction on the proliferation of chondrocytes and the expression of RANKL mRNA (receptor activator nuclear factor kappa B ligand). METHODS: Using serum pharmacology and rabbit chondrocytes cultured in vitro, the effect of serum containing DXHD on chondrocyte proliferation was detected by MTS/PMS system, and the expression of RANKL mRNA in chondrocytes was detected by real-time fluorescent quantitative PCR. RESULTS: The serum containing XZHDI had promoting effect on the proliferation of rabbit articular chondrocytes cultured in vitro. The difference was statistically significant compared with the experimental control group (P<0.05). The RANKL mRNA expression was detected in the serum of the ZIFHTD group. The experimental control group was less than the same time node. Conclusion: The serum containing Zhan Hu Di Decoction can promote the proliferation of chondrocytes cultured in vitro and inhibit the expression of RANKL mRNA in chondrocytes.
