目的:建立和确证一种简便、准确的测定大鼠全血中银浓度的分析方法。方法:选取铟(115In)为内标,先用硝酸对全血样品进行微波消解处理,再采用电感耦合等离子体质谱法测定消化液中银离子的浓度。结果:内源性物质不干扰银和内标的测定,全血中银的浓度在5.0~1 000μg·L-1范围内线性关系良好,日内精密度RSD≤3.3%,日间精密度RSD≤3.8%;提取回收率为97.5%~101.9%,基质效应为97.4%~99.4%。应用该法研究了大鼠静脉注射纳米银后银的体内经时变化,结果表明,给药(40 nm)后,大鼠血液中银浓度先迅速下降而后上升,之后再缓慢消除。结论:该方法可为纳米银在生物机体内的评价研究提供分析方法参考。
Objective: To establish and confirm a simple and accurate method for the determination of silver concentration in rat whole blood. Methods: Indium (115In) was used as the internal standard. The whole blood sample was first treated with nitric acid for microwave digestion, and the concentration of silver ions in the digestion solution was determined by inductively coupled plasma mass spectrometry. Results: The endogenous substances did not interfere with the determination of silver and internal standard. The concentration of silver in whole blood ranged from 5.0 to 1 000 μg · L-1 with a good linearity. The intra-day precision RSD was less than or equal to 3.3% and intra-day precision RSD was less than or equal to 3.8% The extraction recovery was 97.5% -101.9% and the matrix effect was 97.4% -99.4%. The method was used to study the in vivo time-dependent changes of silver nanoparticles after intravenous injection of silver nanoparticles in rats. The results showed that the concentration of silver in the blood of rats dropped rapidly and then slowly after administration of 40 nm, and then slowly disappeared. Conclusion: This method can provide an analytical method reference for the evaluation of nano-silver in biological organisms.
