目的:研究HCV重组体J6/JFH1在Huh7细胞中复制并产生感染性病毒颗粒。方法:将HCV RNA转染至Huh7细胞中,待转染效率达80%左右,收集上清液,感染新的Huh7细胞,得到的病毒命名为一代病毒,取一代病毒感染效率达80%左右的上清液,感染新的Huh7细胞,得到的病毒命名为二代病毒,测定转染与2次感染收集病毒的滴度。结果:在将HCV病毒转染和感染Huh7细胞后, HCV阳性细胞达80%的时间分别是第13天、第5天及第5天,三次最高滴度分别是3.9FFU/ml,4.5FFU/ml及4.6FFU/ml。结论:成功在Huh7细胞内培育出带有适应性突变的J6/JFH1病毒,为HCV体外培育提供了新思路,为耐IFN的丙肝治疗奠定了基础。“,”ObjectiveStudy of HCV recombinant J6 / JFH1 in Huh7 cells copy and produce the infectious virus particles.MethodsHCV RNA transfection to Huh7 cells, after waiting for transfection efficiency up to 80% or so, clear fluid collection, new Huh7 cells of infection, the virus named generation, from a generation of virus infection efficiency about 80% supernatant, new Huh7 cells of infection, the virus named the second generation, determination of transfection with two collected virus drops.ResultsThe HCV virus transfection and Huh7 cells after infection, HCV positive cells reached 80% of the time is 13 days, 5 days and 5 days, three times the highest drop degrees are respectively 3.9 head/ml, 4.5 head/head/ml and 4.6 ml.ConclusionsSuccess in Huh7 cells produced with adaptive mutations J6 / JFH1 virus, for HCV in vitro cultivation provides a new train of thought, which laid a foundation for hepatitis c treatment of IFN resistance.
