目的检测冬凌草甲素对人乳腺癌细胞MDA-MB 231侵袭的影响并探讨其作用的分子机制。方法用Transwell实验检测冬凌草甲素对人乳腺癌细胞MDA-MB 231侵袭的影响;明胶酶谱实验检测冬凌草甲素对基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)酶活性的影响;荧光素酶报告基因实验检测冬凌草甲素对MMP-2和MMP-9启动子活性的影响。结果冬凌草甲素明显抑制MDA-MB 231细胞的侵袭,且明显下调MMP-2和MMP-9的启动子活性和酶活性(P<0.05或<0.01)。结论冬凌草甲素能显著抑制乳腺癌细胞的侵袭,且这种抑制作用可能通过下调MMP-2和MMP-9的启动子活性和酶活性而实现。
Objective To investigate the effect of oridonin on the invasion of human breast cancer cell line MDA-MB 231 and to explore its molecular mechanism. Methods The effect of oridonin on the invasion of human breast cancer cell line MDA-MB 231 was detected by Transwell assay. The effect of oridonin on MMP-2 and MMP- (MMP-9). The luciferase reporter assay was used to detect the effect of oridonin on the activity of MMP-2 and MMP-9 promoter. Results Oridonin significantly inhibited the invasion of MDA-MB-231 cells and significantly down-regulated the promoter activity and enzyme activity of MMP-2 and MMP-9 (P <0.05 or <0.01). Conclusion Oridonin can significantly inhibit the invasion of breast cancer cells, and this inhibition may be achieved by down-regulating the promoter activity and enzyme activity of MMP-2 and MMP-9.
