目的:探讨提高淋巴因子活性细胞(LAK细胞)单个细胞和总体的抗瘤活性。方法:对99例晚期肿瘤患者进行自体LAK细胞培养,取患者抗凝(肝素)外周血,加入淋巴细胞分层液离心,取得外周血单个核细胞,然后按一定的标准放于37℃、5%CO2环境中培养、扩增、回输。结果:分析得出白细胞介素2、CD3单抗、红细胞、植物凝血素可促进LAK细胞增殖及提高其活性。适当的化疗对LAK细胞的培养无明显影响。结论:LAK细胞回输的最佳时间是7~15d。
Objective: To investigate the anti-tumor activity of single cells of lymphokine-activated cells (LAK cells) and their overall enhancement. Methods: Ninety-nine patients with advanced cancer were cultured with autologous LAK cells. Peripheral blood of patients with anticoagulant (heparin) was taken out and centrifuged by adding lymphocyte stratified liquid. Peripheral blood mononuclear cells were obtained. The cells were then stored at 37 ℃, 5 % CO2 environment culture, amplification, back to lose. RESULTS: Interleukin 2, CD3 monoclonal antibody, erythrocyte and lectin were found to promote LAK cell proliferation and increase their activity. Appropriate chemotherapy has no obvious effect on the culture of LAK cells. Conclusion: The optimal time for LAK cell reinfusion is 7-15 days.
