目的:探讨四妙勇安汤对实验性腹主动脉瘤(Abdominal Aortic Aneurysm,AAA)生长的影响及机制。方法:建立大鼠AAA模型,随机分成四妙勇安汤组和盐水对照组,造模后2周、4周,应用弹力纤维染色、CD68免疫组织化学染色观察动脉壁弹力纤维的损伤、巨噬细胞的浸润,用RT-PCR和Western蛋白质印迹方法观察单核细胞趋化蛋白-1(monocyte chemotacticprotein-1,MCP-1)及基质金属蛋白酶-2(matrix metalloproteinases-2,MMP-2)的基因表达。结果:与盐水对照组相比,造模后2周时,四妙勇安汤组大鼠腹主动脉的扩张、动脉壁弹力纤维的损伤、CD68阳性表达、MCP-1和MMP-2的基因表达程度均明显降低(P<0.01)。结论:四妙勇安汤能通过抑制动脉壁炎性细胞的浸润和相关基因的表达,对实验性腹主动脉瘤的生长产生明显的抑制作用。
Objective: To investigate the effect and mechanism of Si Miao Yong An Tang on the growth of experimental abdominal aortic aneurysm (AAA). METHODS: Rat AAA model was established and randomly divided into SJM group and saline control group. After 2 weeks and 4 weeks after modeling, elastic fiber staining and CD68 immunohistochemical staining were used to observe the damage and macrophages of the arterial wall elastic fibers. Infiltration of cells, monocyte chemotactic protein-1 (MCP-1) and matrix metalloproteinase-2 (MMP-2) genes were observed by RT-PCR and Western blot. expression. RESULTS: Compared with the saline control group, at the 2 weeks after the modeling, abdominal aorta dilation, damage of arterial wall elastic fibers, CD68 positive expression, MCP-1 and MMP-2 gene expression were observed in the rats of the SMM group. The degree of expression was significantly reduced (P<0.01). Conclusion: Si Miao Yong An Decoction can inhibit the growth of experimental abdominal aortic aneurysms by inhibiting the infiltration of inflammatory cells in the arterial wall and the expression of related genes.
