目的观察鳖甲煎丸对肾间质纤维化实验大鼠肾脏的保护作用并探讨其作用机理。方法采用单侧输尿管结扎的方法复制大鼠肾梗阻模型,将其随机分为缬沙坦组、鳖甲煎丸组与模型组,并设假手术组。观察梗阻侧肾脏肾上腺髓质素(ADM)在蛋白及基因水平的表达,并测定其在肾组织中蛋白含量。结果模型组及各治疗组大鼠肾脏中肾上腺髓质素组织蛋白含量、蛋白表达以及mRNA的基因表达较假手术组明显降低,鳖甲煎丸组、缬沙坦组的含量及表达均高于模型组(P<0.05?,但两组之间比较无显著性差异。结论鳖甲煎丸能够明显上调肾间质纤维化大鼠肾脏ADM的蛋白及mRNA的表达,对肾脏起到保护作用。
Objective To observe the protective effect of Biejiajianwan on kidney of rats with renal interstitial fibrosis and explore its mechanism of action. Methods Rat kidney obstruction models were made by unilateral ureteral ligation. They were randomly divided into valsartan group, Biejiajianwan group and model group, and sham operation group was set up. The expression of adrenomedullin (ADM) in the obstructed side of the kidney was observed at the protein and gene level, and the protein content in the renal tissue was determined. Results Compared with the sham operation group, the content of adrenomedullin, protein expression and mRNA expression of the kidneys in the model group and the treatment group rats were significantly lower than those in the sham operation group. The contents and expression of the sham group and valsartan group were higher than those in the sham operation group. The model group (P<0.05), but there was no significant difference between the two groups. Conclusion Biejiajianwan can significantly up-regulate the protein and mRNA expression of renal ADM in rats with renal interstitial fibrosis, and protect the kidney.
