目的 :研究异甘草素对人胰腺癌SW1990细胞增殖及细胞内survivin表达的影响。方法 :体外培养SW1990细胞,给予不同浓度的异甘草素处理,MTT法检测异甘草素对SW1990细胞增殖的影响,实时定量PCR技术检测肿瘤细胞survivin基因m RNA的表达,蛋白免疫印迹法检测survivin蛋白的表达。结果:在一定浓度范围内,异甘草素能显著抑制人胰腺癌SW1990细胞的增殖,并呈时间与剂量依赖性,且异甘草素能显著抑制人胰腺癌SW1990细胞内survivin基因m RNA及蛋白的表达。结论:异甘草素对人胰腺癌SW1990细胞增殖有明显的抑制作用,且可能与下调survivin基因m RNA及蛋白的表达有关。
Objective: To investigate the effect of isoliquiritigenin on the proliferation and the expression of survivin in human pancreatic cancer cell line SW1990. Methods: SW1990 cells were cultured in vitro and treated with different concentrations of isoliquiritigenin. The effects of isoliquiritigenin on the proliferation of SW1990 cells were detected by MTT assay. The expression of survivin mRNA was detected by real-time quantitative PCR. The expression of survivin protein expression. Results: Isoliquiritigenin could significantly inhibit the proliferation of human pancreatic cancer cell line SW1990 in a time-and dose-dependent manner, and isoliquiritigenin could significantly inhibit the expression of survivin m RNA and protein in human pancreatic cancer SW1990 cells expression. CONCLUSION: Isoliquiritigenin can significantly inhibit the proliferation of human pancreatic cancer cell line SW1990 and may be related to the down-regulation of m RNA and protein expression of survivin gene.
