目的探讨胞外钙离子对HepG2细胞和LO2细胞的细胞毒性以及与阿霉素联合对两种细胞增殖抑制作用的影响。方法采用四甲基偶氮唑蓝法检测细胞活力,流式细胞术检测细胞凋亡率和坏死率。结果各个剂量组,胞外钙离子对HepG2细胞的增殖抑制作用均高于LO2细胞(均有P<0.05),HepG2细胞的坏死率和凋亡率都高于LO2细胞(均有P<0.05)。胞外钙离子与阿霉素联合作用能选择性增强阿霉素诱导HepG2细胞坏死,且表现出交互效应(P<0.001),但对LO2细胞坏死无影响。结论胞外钙离子对HepG2细胞更敏感,细胞增殖抑制作用高于LO2细胞,能显著提高阿霉素对HepG2细胞的杀灭作用。
Objective To investigate the cytotoxicity of extracellular calcium to HepG2 cells and LO2 cells and the effect of combined with doxorubicin on the proliferation of both cells. Methods Cell viability was detected by MTT method. Apoptosis rate and necrosis rate were detected by flow cytometry. Results The proliferation of HepG2 cells was significantly higher than that of LO2 cells (all P <0.05). The necrosis and apoptosis rates of HepG2 cells were higher than those of LO2 cells (all P <0.05) . Extracellular calcium combined with doxorubicin selectively enhanced doxorubicin-induced HepG2 cell necrosis and showed an interactive effect (P <0.001), but had no effect on LO2 cell necrosis. Conclusion Extracellular calcium ion is more sensitive to HepG2 cells and has higher cell proliferation inhibition than LO2 cells, which can significantly increase the killing effect of doxorubicin on HepG2 cells.
