①目的探讨阿霉素对抗阿霉素细胞系S-180-R细胞凋亡的作用。②方法用低剂量(3.25μg)和高剂量(6.50μg)阿霉素分别处理亲本S-180和抗药S-180-R细胞腹腔接种的BABL/C鼠,72h取腹水,用流式细胞仪测定细胞DNA相对含量。③结果低剂量阿霉素72h使亲本S-180细胞出现62.5%的凋亡,使抗药S-180-R仅出现13.0%的凋亡细胞,高剂量阿霉素72h使亲本S-180细胞几乎全部死亡,使S-180-R出现20.0%的细胞凋亡。低剂量阿霉素处理72h,亲本S-180细胞鼠腹腔出现大量淋巴细胞;抗药S-180-R细胞鼠腹腔只有少量淋巴细胞出现。高剂量阿霉素处理亲本S-180细胞仍保留很多淋巴细胞,其他S-180细胞几乎全部死亡,而抗药S-180-R细胞中淋巴细胞消失。④结论细胞抗凋亡是细胞抗药性的明显标志之一。抗阿霉素S-180-R细胞接种的小鼠腹腔内淋巴细胞的数量与阿霉素的剂量有关。
Objective To investigate the effect of doxorubicin on the apoptosis of adriamycin cell line S-180-R. 2Methods BABL/C mice were inoculated intraperitoneally with parental S-180 and drug-resistant S-180-R cells at low dose (3.25 μg) and high dose (6.50 μg) doxorubicin respectively. Ascites fluid was used for 72 hours. Cytometry was used to determine the relative cellular DNA content. 3 Results Low-dose doxorubicin 72h caused 62.5% apoptosis in parental S-180 cells, making anti-drug S-180-R only 13.0% of apoptotic cells, high-dose doxorubicin 72h to make the parent Almost all of the S-180 cells died, causing apoptosis of S-180-R by 20.0%. Low-dose doxorubicin treatment 72h, a large number of lymphocytes in the abdominal cavity of the parent S-180 cells; only a small amount of lymphocytes appeared in the abdominal cavity of mice resistant to S-180-R cells. High-dose doxorubicin-treated parental S-180 cells still retained many lymphocytes, almost all of the other S-180 cells died, and lymphocytes disappeared in the resistant S-180-R cells. 4 Conclusion The anti-apoptosis of cells is one of the obvious signs of cell resistance. The number of intraperitoneal lymphocytes in mice inoculated with doxorubicin S-180-R cells was related to the dose of doxorubicin.
