目的探讨大豆异黄酮(ISF)联合长春瑞滨(NVB)化疗对A549肺癌细胞增殖影响。方法采用噻唑兰法观察ISF和NVB联用对A549肺癌细胞增殖影响,采用Weeb系数法判定联合用药相互作用,同时采用透射电镜检测A549细胞形态学变化。结果 ISF和NVB单独使用均可时间和浓度依赖性地抑制肺癌A549细胞增殖;20~80 mg/L ISF作用48 h,A549细胞增殖抑制率为15%~23%;8~32 mg/L NVB作用48 h,A549细胞增殖抑制率为53%~80%;ISF浓度在40 mg/L时,ISF和NVB联用后抗A549细胞增殖作用增强,对肺癌细胞生长抑制作用呈量效关系;且NVB浓度在8~32 mg/L时,抑制率为71%~93%,二者有相加或协同作用。结论 ISF和NVB联用可增强对肺癌A549细胞增殖抑制,二者具有协同作用。
Objective To investigate the effects of isoflavone (ISF) plus vinorelbine (NVB) chemotherapy on the proliferation of A549 lung cancer cells. Methods The effect of combination of ISF and NVB on the proliferation of A549 lung cancer cells was observed by thiazolyl method. Weeb coefficient method was used to determine the effect of combination therapy. Morphological changes of A549 cells were detected by transmission electron microscopy. Results Both ISF and NVB alone could inhibit the proliferation of lung cancer A549 cells in a time and concentration dependent manner. Inhibition rates of proliferation of A549 cells treated with 20-80 mg / L ISF for 48 h were 15% -23% The inhibitory rate of proliferation of A549 cells was 53% ~ 80% at 48 h. When the concentration of ISF was 40 mg / L, the proliferation of A549 cells was enhanced by combination of ISF and NVB, When the concentration of NVB was 8 ~ 32 mg / L, the inhibition rate was 71% -93%, and the two had additive or synergistic effect. Conclusions Combination of ISF and NVB can inhibit the proliferation of lung cancer cell line A549 with synergistic effect.
