以马铃薯中α-茄碱为研究对象,研究不同发芽天数、芽周围不同取样深度马铃薯中α-茄碱含量变化规律。样品经5%乙酸水振荡提取,4℃低温离心取上清液,采用超高效液相色谱串联质谱法测定,质谱采用多反应监测、正离子扫描模式,分别测定马铃薯皮和肉中α-茄碱含量。结果表明,在0、4、6、8、10 d获取的马铃薯皮和肉样品,α-茄碱均随时间呈上升趋势,且马铃薯皮中α-茄碱含量远高于肉。对于发芽的马铃薯,距离皮越远的肉中α-茄碱含量越低。所有马铃薯样品肉中含量均低于200 mg/kg,都在安全范围内,削掉芽和皮后食用,理论上不会对人体健康造成危害。
Taking α-solanine in potato as the research object, the variation regularity of α-solanine content in potatoes with different germination days and buds was studied. Samples were extracted with 5% acetic acid in water and centrifuged at 4 ℃. The supernatants were separated by ultra performance liquid chromatography-tandem mass spectrometry. Multivariate reaction monitoring and positive ion scanning were used for the determination of α- Alkali content. The results showed that the samples of potato husk and meat, α-solanine, which were obtained at 0, 4, 6, 8 and 10 d, all showed an upward trend with time, and the content of α-solanine in potato husks was much higher than that of meat. For germinated potatoes, the farther away the skin, the lower the alpha-solanine content. All potato samples contain less than 200 mg / kg of meat, are within safe limits, cut off the buds and skin after eating, in theory, will not cause harm to human health.
