目的为进一步研究雌激素受体β亚型(ERβ)对小鼠睾丸发育及性功能调节的作用,观察了出生后1,2,3,4周的小鼠睾丸和成年小鼠睾丸中ERβ的表达和分布情况。方法采用出生1,2,3,4周雄性昆明小鼠的发育过程以及成年雄性昆明小鼠各2只,分成2组,第1组4只,第2组4只。采用免疫组织化学染色和免疫荧光组织化学染色方法检测小鼠睾丸中ERβ。结果小鼠睾丸中ERβ的表达随出生时间增长而呈逐渐上升的趋势,在出生后1,2周的小鼠睾丸中ERβ主要分布于睾丸间质细胞,在出生后3,4周小鼠睾丸中ERβ分布于睾丸间质细胞胞质和细胞核,核仁呈阴性,成年小鼠睾丸间质细胞中ERβ分布较幼年小鼠密度增加。结论ERβ在小鼠睾丸发育过程中可能作为一种特异性受体通过睾丸间质细胞影响和调节雄性激素的分泌进而对生精过程和精子的成熟有重要的调节作用。
Objective To further study the effect of estrogen receptor β subtype (ERβ) on testicular development and sexual function in mice, we observed the changes of ERβ Expression and distribution. Methods The development of male Kunming mice at 1, 2, 3 and 4 weeks of birth and 2 adult male Kunming mice were divided into 2 groups, 4 in group 1 and 4 in group 2 respectively. Immunohistochemical staining and immunofluorescence staining were used to detect ERβ in mouse testes. Results The expression of ERβ in mouse testis tended to increase gradually with birth time. ERβ mainly distributed in testicular stromal cells in testis of mice 1 and 2 weeks after birth, ERβ was distributed in the cytoplasm and nucleus of testicular stromal cells. The nuclei were negative. The distribution of ERβ in testicular stromal cells of adult mice increased compared with that of young mice. Conclusion ERβ may play an important role in the process of spermatogenesis and sperm maturation as a specific receptor through the influence of testicular stromal cells and the regulation of androgen secretion in mouse testes.
