目的:了解大肠癌组织中MAD2和p53表达概况,探寻其表达差异及临床意义。方法:通过实时荧光定量PCR和免疫组织化学方法检测大肠癌、腺瘤及正常大肠黏膜组织中MAD2表达,并对大肠癌组织中MAD2扩增产物进行测序。同时利用免疫组织化学方法检测大肠癌和正常组织中p53的表达。结果:大肠癌组织中MAD2的阳性表达明显高于腺瘤和正常大肠黏膜组织,三者比较差异有统计学意义,P<0.001。MAD2表达与分化程度和生存时间有关,P<0.05。大肠癌组织中未见MAD2基因突变。正常大肠黏膜组织中p53表达均为阴性,大肠癌组织中p53阳性表达率为50%。结论:MAD2和p53表达异常是大肠癌产生的机制之一,与大肠癌的发生、发展有关。
Objective: To investigate the expression of MAD2 and p53 in colorectal carcinoma and explore their differences and clinical significance. Methods: The expression of MAD2 in colorectal carcinoma, adenoma and normal colorectal mucosa was detected by real-time fluorescence quantitative PCR and immunohistochemistry. The MAD2 amplification product in colorectal cancer tissues was sequenced. At the same time, immunohistochemistry was used to detect the expression of p53 in colorectal cancer and normal tissues. Results: The positive expression of MAD2 in colorectal carcinoma was significantly higher than that in adenoma and normal mucosa, the difference was statistically significant (P <0.001). MAD2 expression and the degree of differentiation and survival time, P <0.05. No MAD2 gene mutation in colorectal cancer tissues. Normal colorectal mucosa p53 expression were negative, colorectal cancer p53 positive expression rate of 50%. Conclusion: The abnormal expression of MAD2 and p53 is one of the mechanisms of colorectal cancer, which is related to the occurrence and development of colorectal cancer.
