目的探讨哮喘模型大鼠肺泡巨噬细胞(alveolar macrophage,AM)电压依赖性钾通道(Voltage-dependent potassiumchannel,KV)活性的变化及其对巨噬细胞功能的影响。方法利用卵蛋白(ovialbumin,OVA)致敏并激发大鼠建立哮喘模型,收集支气管肺泡灌洗液(bronchus alveolarlavagefluid,BALF)培养AM。利用全细胞电压钳模式记录AM细胞膜上的KV电流,观察比较哮喘模型组和对照组KV通道活性的变化。结果哮喘组支气管肺泡灌洗液(BALF)细胞总数显著增高,与对照组(n=7)比较差异显著(P<0.01);哮喘大鼠AM细胞KV电流幅度[(243.56±133.56)pA,n=11]较对照组[(656.23±162.34)pA,n=11]明显降低(P<0.01)。结论哮喘大鼠AM细胞KV通道活性降低,可能导致AM兴奋性增高易被激活分泌各种炎性介质和细胞因子,从而参与哮喘炎症的形成。
Objective To investigate the changes of voltage-dependent potassium channel (KV) activity in alveolar macrophages (AM) and its effect on the function of macrophages in asthmatic rats. Methods The asthmatic model was established by ovargbumin (OVA) sensitization and challenge in rats. Bronchoalveolar lavage fluid (BALF) was collected to culture AM. KV current on AM cell membrane was recorded by whole-cell voltage-clamp mode, and the changes of KV channel activity in asthmatic model group and control group were observed and compared. Results The total number of bronchoalveolar lavage fluid (BALF) cells in asthma group was significantly higher than that in control group (n = 7) (P <0.01). The KV current amplitude of AM cells in asthmatic rats [(243.56 ± 133.56) pA, n = 11] was significantly lower than that of the control group [(656.23 ± 162.34) pA, n = 11] (P <0.01). Conclusion The decrease of KV channel activity of AM cells in asthmatic rats may lead to the increase of AM excitability. It is easy to be activated to secrete various inflammatory mediators and cytokines and thus participate in the formation of asthmatic inflammation.
