目的研究榄香烯乳对人胰腺癌细胞株Panc-1增殖、凋亡的作用及可能机制。方法不同剂量榄香烯乳0、50、100、150、200、250μg/ml诱导Panc-1细胞后,采用MTT法检测细胞体外生长情况,流式细胞术分析细胞周期及细胞凋亡率,实时荧光定量PCR检测B细胞淋巴瘤-白血病2(Bcl-2)、半胱天冬氨酸蛋白酶3(Caspase-3)的表达。结果榄香烯乳呈剂量依赖性地抑制Panc-1细胞增殖,降低G0/G1期细胞比例和Bcl-2表达,增加S期细胞比例、细胞凋亡率及Caspase-3表达。结论榄香烯乳抑制Panc-1细胞增殖,诱导细胞凋亡;这可能与Bcl-2低表达及Caspase-3高表达有关。
Objective To study the effects of elemene on proliferation and apoptosis of human pancreatic cancer cell line Panc-1 and its possible mechanism. Methods Panc-1 cells were induced by different doses of elemene at 0, 50, 100, 150, 200 and 250μg / ml. The growth of Panc-1 cells was detected by MTT assay. Cell cycle and apoptosis rate were analyzed by flow cytometry. Fluorescent quantitative PCR was used to detect the expression of Bcl-2 and Caspase-3. Results Elemene inhibited the proliferation of Panc-1 cells in a dose-dependent manner, and decreased the percentage of cells in G0 / G1 phase and the expression of Bcl-2, increased the proportion of cells in S phase, the rate of apoptosis and the expression of Caspase-3. Conclusion Elemene inhibited the proliferation and induced apoptosis of Panc-1 cells, which may be related to the low expression of Bcl-2 and the high expression of Caspase-3.
