Toll样受体4及相关炎性信号分子在高脂饮食诱导的肥胖小鼠肾脏组织水平升高

目的探讨Toll样受体4(TLR4)、髓样分化因子88(My D88)、核因子κBp65(NF-κBp65)在高脂饮食(HFD)诱导的肥胖小鼠肾脏组织中的表达。方法 45只健康C57BL/6雄性小鼠随机分为HFD组(n=39)和对照组(n=6)。HFD组给予高脂高糖饲料,对照组给予普通饲料,均喂养20周。每周测量小鼠体质量,取体质量增长值最高的13只为饮食诱导肥胖(DIO)组,称量内脏脂肪质量。检测血清肌酐、尿素氮(BUN)及内毒素水平。HE染色、显微镜观察肾脏形态。实时定量PCR(qRT-PCR)检测小鼠肾脏组织TLR2、TLR4、My D88、NF-κBp65及TNF-αmRNA水平,Western blot法检测小鼠肾脏组织TLR4、My D88、NF-κBp65蛋白水平。结果成功构建DIO小鼠模型;与对照组相比,DIO小鼠血内毒素水平升高,血肌酐、BUN水平无显著性差异,2组小鼠肾脏结构未见明显异常;与对照组小鼠相比,DIO小鼠肾脏TLR4、My D88、NF-κBp65、TNF-αmRNA水平升高,TLR2 mRNA水平无显著性差异;DIO小鼠肾脏TLR4、My D88、NF-κBp65蛋白水平升高。结论 TLR4及相关炎性信号分子在饮食诱导肥胖小鼠肾脏组织表达水平增加。 Objective To investigate the expression of TLR4, My D88 and NF-κBp65 in obese mice induced by high fat diet (HFD). Methods 45 healthy C57BL / 6 male mice were randomly divided into HFD group (n = 39) and control group (n = 6). HFD group were given high-fat and high-sugar diet, the control group were given normal feed, were fed for 20 weeks. The body weight of mice was measured weekly, and the highest value of body weight gain was taken from the diet-induced obesity (DIO) group and the visceral fat mass was weighed. Serum creatinine, blood urea nitrogen (BUN) and endotoxin levels were measured. HE staining, microscopic observation of kidney morphology. The levels of TLR2, TLR4, My D88, NF-κBp65 and TNF-αmRNA were detected by real-time quantitative PCR (qRT-PCR) in kidney and the protein levels of TLR4, My D88 and NF-κBp65 in kidney were detected by Western blot. Results Compared with the control group, the level of blood endotoxin in DIO mice increased and the levels of serum creatinine and BUN were not significantly different. The structure of kidney in the two groups showed no obvious abnormality. Compared with the control group Compared with the control group, the levels of TLR4, My D88, NF-κBp65 and TNF-α mRNA in the kidney of DIO mice did not change significantly. The protein levels of TLR4, My D88 and NF-κB p65 in DIO mice kidney were increased. Conclusion The expression of TLR4 and related inflammatory signaling molecules in kidney increased in diet-induced obese mice.
