目的:为放射免疫显像在诊断食管癌淋巴结转移中的应用,提供基础研究支持。方法:①免疫组化(LSAB法)测定不同组织冷冻切片和抗人食管鳞癌单抗G9的反应;②125I标记G9,形成标记化合物,在荷食管癌裸鼠腹腔注射125I-G9后的连续3天内,测定各主要组织、器官的放射性。结果:①LSAB显示,食管癌原发灶和转移淋巴结呈阳性反应,正常食管和无转移淋巴结阴性。②125I-G9在裸鼠体内分布显示,肿瘤组织的放射性计数均明显高于其它器官/组织;各器官/组织的T/NT值在2~7之间;放射自显影的结果也显示,肿瘤部位有大量的放射性浓集。结论:①单抗G9能选择性定位于食管癌原发灶与转移淋巴结的癌细胞胞膜。②125I-G9在荷食管癌裸鼠有肿瘤组织导向作用,显像效果良好,提示放射免疫显像在食管癌转移淋巴结定位,有潜在的临床应用前景。
Objective: To provide basic research support for the application of radioimmunoimaging in the diagnosis of lymph node metastasis of esophageal cancer. Conclusion: ① McAb G9 can selectively locate in the esophageal cancer metastasis and lymph node metastasis of cancer cell membrane. ②125I-G9 in tumor-bearing nude mice with tumor-oriented role, imaging effect is good, suggesting that radioimmunoimaging metastasis in esophageal cancer lymph node mapping, there is potential for clinical application.
