目的研究雷公藤多甙对类风湿关节炎滑膜产生趋化因子RANTES、MCP-1的影响。方法取经雷公藤多甙、MTX治疗后的CIA大鼠关节滑膜组织,提取总RNA,用RT-PCR法测定其中RANTESmRNA、MCP-1mRNA水平。结果雷公藤多甙能抑制滑膜组织中趋化因子MCP-1mRNA、RANTESmRNA的表达,MTX对MCP-1mRNA、RANTESmRNA的影响作用不明显。结论雷公藤多甙可抑制类风关滑膜趋化因子RANTES、MCP-1产生。
Objective To study the effects of Tripterygium wilfordii on chemokines RANTES and MCP-1 produced by synovial membranes in rheumatoid arthritis. Methods The synovial tissues of CIA rats treated with Tripterygium wilfordii Hook and MTX were used to extract total RNA and the levels of RANTES mRNA and MCP-1 mRNA were determined by RT-PCR. RESULTS: Tripterygium wilfordii could inhibit the expression of chemokine MCP-1 mRNA and RANTES mRNA in synovial tissue. MTX had no obvious effect on MCP-1 mRNA and RANTES mRNA. Conclusion Tripterygium wilfordii can inhibit the production of RANTES and MCP-1, the chemokine of wind-like synoviocytes.
