本文采用区带密度梯度离心,国内首次分离得到黄牛纯化的高密度脂蛋白(HDL)。然后用Sephadex G-200凝胶过滤,可将其中的载脂蛋白分为3个组分。通过聚丙烯酰胺凝胶电泳分析,确认组分Ⅱ为载脂蛋白A-Ⅰ(ApoA-Ⅰ)占HDL的80~90%。测定其分子量为27040道尔顿,整个氨基酸(AA)组成中不含半胱氨酸,其N端是天冬氨酸。本文确认了黄牛是含ApoA-Ⅰ丰富的动物之一。同时也为分离、提纯HDL和ApoA-Ⅰ提供了可靠易行的方法。
In this study, density-gradient centrifugation was used in the study. The purified HDL was isolated from cattle for the first time in China. Then, the apolipoprotein was separated into 3 fractions by Sephadex G-200 gel filtration. By polyacrylamide gel electrophoresis analysis, it is confirmed that component II is apolipoprotein A-I (ApoA-I) accounting for 80-90% of HDL. Its molecular weight was determined to be 27040 daltons. The entire amino acid (AA) composition did not contain cysteine and the N-terminal end was aspartic acid. This study confirmed that cattle are one of the most abundant animals that contain ApoA-I. It also provides a reliable and easy way to separate and purify HDL and ApoA-Ⅰ.
