目的观察给以不同剂量氟的大鼠胰岛素水平的变化。方法经灌胃给氟复制氟中毒模型。利用Siemens ADVIA Centaur XP全自动化学发光免疫分析仪检测大鼠血清胰岛素水平,酶联免疫吸附法分析糖化血红蛋白水平,使用贝克曼全自动生化分析仪检测血脂。结果胰岛素和糖化血红蛋白水平的变化显示出低剂量和高剂量的氟都能够诱导胰岛素分泌,其高剂量氟组大鼠胰岛素水平更高。高剂量氟对胰岛素耐受影响不大,而低剂氟组大鼠胰岛素耐受能力下降,联合胰岛功能受损,进一步降低了大鼠对胰岛素的敏感性。结论一定剂量的氟刺激大鼠胰岛素的分泌,且对胰岛素的分泌成剂量-效应关系。此外,氟能够影响大鼠对胰岛素的敏感性,甚至降低其敏感性。
Objective To observe the changes of insulin levels in rats given different doses of fluoride. Methods Fluoride poisoning model was established by intragastric administration of fluoride. Serum insulin levels were measured using a Siemens ADVIA Centaur XP automated chemiluminescence immunoassay analyzer, and HbA1c levels were analyzed by enzyme-linked immunosorbent assay (ELISA). Lipid levels were measured using a Beckman automated biochemical analyzer. Results The changes of insulin and glycosylated hemoglobin showed that both low dose and high dose of fluoride could induce insulin secretion, and insulin in high dose fluoride group was higher. High dose of fluoride has little effect on insulin resistance, while low dose of fluoride rats decreased insulin resistance, combined with islet dysfunction, further reducing the sensitivity of rats to insulin. Conclusion A certain dose of fluoride stimulates the secretion of insulin in rats and has a dose-effect relationship with insulin secretion. In addition, fluoride can affect rats’ insulin sensitivity and even reduce their sensitivity.
