环孢菌素A是一种强效的免疫抑制剂,临床上用于治疗器官排斥反应,也可用于治疗某些自身免疫性疾病。本文报道一种测定全血中环孢菌素A的HPLC方法,该法适合于实验室常规监测。色谱系统色谱系统由Waters 510型泵、712 WISP自动进样器、TCM柱加热器、490 E可变波长紫外检测器及Waters 820型色谱工作台组成。反相Velosep BrownleeRP 18色谱柱,4cm×3.2mm,粒度3μm。在泵与自动进样器之间安装一个C18预饱和柱。分析柱和预饱和柱的柱温均控制在70℃。流动相由50%乙腈、11%甲醇和39%0.01M磷酸氢二钾缓冲液(pH6.5)三种
Cyclosporin A is a potent immunosuppressive agent that is clinically used to treat organ rejection and can also be used to treat certain autoimmune diseases. This article reports an HPLC method for the determination of cyclosporine A in whole blood, which is suitable for routine laboratory monitoring. Chromatography The system consists of a Waters 510 pump, a 712 WISP autosampler, a TCM column heater, a 490 E variable wavelength UV detector, and a Waters 820 chromatography bench. Inverse Velosep Brownlee RP 18 column, 4 cm × 3.2 mm, particle size 3 μm. Install a C18 prepacked column between the pump and the autosampler. The analytical column and pre-saturated column temperature were controlled at 70 ℃. The mobile phase consisted of 50% acetonitrile, 11% methanol and 39% 0.01 M dipotassium hydrogen phosphate buffer (pH 6.5)
