本实验对大鼠下丘脑生长抑素神经元染色,以5%丙烯醛固定,1 mm~3组织块浸泡2.5小时,入30%蔗糖过夜,冰冻或振荡切片机切片50μm后分别用高碘酸钠、氢硼化钠处理,以去除多余的固定剂,后常规漂浮PAP染色。电镜标本用振荡切片机切片,采用平板包理技术,LKB超薄切片,H-800电镜观察。本实验同时对比了Bouin和多聚甲醛固定剂灌注后PAP染色,并进一步观察了三种条件下对LHRH和NT的PAP染色,结果表明,丙烯醛明显优越于其它二种固定剂,它能很好的显示下丘脑各区的阳性细胞和纤维,尤其是细的纤维,并且背底浅淡。电镜观察结果也进一步证明丙烯醛在较好的保存细微结构的同时,比其它任何一种固定液都显示出更强的
In this study, rat hypothalamic somatostatin neurons stained with 5% acrolein fixed, 1 mm ~ 3 tissue block soaked 2.5 hours, 30% sucrose overnight, frozen or vibrated slice slicer 50μm, respectively, with periodic acid Sodium, sodium borohydride treatment to remove excess fixative, conventional floating PAP staining. Electron microscopy specimens were sliced with an oscillating microtome using flat-pack technique, LKB ultrathin sections and H-800 electron microscopy. In the present study, PAP staining of Bouin and paraformaldehyde fixatives was also compared. PAP staining of LHRH and NT was further observed under three conditions. The results showed that acrolein was significantly superior to the other two fixatives, Good to show the various regions of the hypothalamus positive cells and fibers, especially fine fibers, and shallow back. Electron microscopy observations also further prove that acrolein shows better microstructure than any other fixative
