目的 为研究胃癌细胞DNA倍体和细胞周期各时相分布与肿瘤细胞化疗药物敏感性的关系。方法 应用流式细胞仪检测了 1 0 9例胃癌细胞的DNA倍体和细胞周期 ,同时用体外细胞培养和MTT比色法测定肿瘤细胞对 8种化疗药物的敏感性。结果 胃癌细胞异倍体的检出率为 41 .3 % (45/ 1 0 9)。正倍体癌细胞的化疗药物敏感率明显高于异倍体细胞 ,特别以DDP、MMC、VP1 6 明显增高 (P <0 .0 5、P <0 .0 1 ) ;PI增高的增殖性癌细胞的化疗药物敏感率以 5 FU、MTX、VCR、VP1 6 高于非增殖性肿瘤细胞 ,且 5 FU、MTX明显增高 (P <0 .0 5、P <0 .0 1 ) ,而ADM、DDP、CTX敏感率明显低于非增殖细胞 (均P <0 .0 5)。结论 胃癌异倍体细胞的化疗药物敏感率低于正常倍体细胞 ,增殖性癌细胞对周期药物的敏感率高于非增殖性细胞 ,而非周期药物的敏感率降低
Objective To study the relationship between DNA ploidy and cell cycle distribution of gastric cancer cells and chemosensitivity of tumor cells. Methods DNA ploidy and cell cycle of 109 gastric cancer cells were detected by flow cytometry. Meanwhile, the sensitivity of tumor cells to 8 chemotherapeutic agents was determined by in vitro cell culture and MTT colorimetry. Results The detection rate of aneuploidy in gastric cancer cells was 41.3% (45/109). The positive rate of chemotherapeutic drugs in euploid cancer cells was significantly higher than that in aneuploid cells, especially in DDP, MMC and VP1 6 (P <0.05, P <0.01) The chemosensitivity rate of cells to chemotherapy was higher than that of non-proliferative cells with 5 FU, MTX, VCR and VP1 6, and 5 FU and MTX were significantly increased (P <0.05, P <0.01) DDP, CTX sensitivity was significantly lower than non-proliferating cells (P <0.05). Conclusions The sensitivity of chemotherapeutic drugs to gastric aneuploid cells is lower than that of normal ploidy cells, the sensitivity of proliferative cancer cells to periodic drugs is higher than that of non-proliferative cells, and the sensitivity of non-cyclopharmaceuticals is reduced
