目的:建立一株抗人单核-巨噬细胞及中性粒细胞的杂交瘤MAb7.方法:用人肝癌组织细胞悬液免疫BALB/c小鼠,取脾细胞与SP2/0细胞融合,ABC染色法筛选单克隆抗体,APAAP法鉴定髓细胞特征。结果:抗体亚类为小鼠IgGl,相应抗原经亲和层析纯化,Mr60000.免疫组化染色,在石蜡切片的组织中和在血片及骨髓涂片中单核巨噬细胞和成熟的中性粒细胞阳性,单核细胞性白血病及中性粒细胞性白血病阳性,而上皮、内皮、网状组织细胞、成纤维细胞、平滑肌细胞及其它粒细胞、淋巴细胞、红细胞均为阴性.结论:MAb7在研究单核-巨噬细胞的分化、免疫功能及病理分型等方面有一定的意义和价值.
OBJECTIVE: To establish a monoclonal antibody against human monocyte-macrophage and neutrophil hybridoma MAb7. Methods: BALB / c mice were immunized with human hepatocellular carcinoma cell suspension. The spleen cells were fused with SP2 / 0 cells. The monoclonal antibodies were screened by ABC staining and the myeloid cells were identified by APAAP. Results: The antibody subclass was mouse IgG1, and the corresponding antigen was purified by affinity chromatography, Mr60000. Immunohistochemical staining showed that mononuclear macrophages and mature neutrophil-positive, monocytic leukemia and neutrophilic leukemia were positive in tissues of paraffin sections and in blood and bone marrow smears, while epithelial , Endothelium, reticular tissue cells, fibroblasts, smooth muscle cells and other granulocytes, lymphocytes, red blood cells were negative. Conclusion: MAb7 has certain significance and value in studying the differentiation of monocyte - macrophage, immune function and pathological typing.
