目的:研究智生胶囊I号方(ZSJN)对老年痴呆模型大鼠智力、单胺氧化酶(MAO)、β-淀粉样蛋白(β-AP)、谷胱甘肽过氧化物酶(GSH-Px)的影响,探讨其防治老年痴呆的作用及机理。方法:应用腹腔注射D-半乳糖制成老年痴呆大鼠,应用Morris水迷宫法判断大鼠的学习记忆能力,采用生化及其放免法测定其脑内MAO活性、β-AP及GSH-Px含量。结果:与正常组大鼠相比,模型组学习记忆能力明显降低。应用“智生胶囊I号方”预防和治疗后,模型鼠学习记忆能力得到保护和改善,脑组织MAO活性降低、β-AP含量减少、GSH-Px酶升高。结论:智生胶囊I号方能明显增强模型大鼠学习记忆能力,通过降低单胺氧化酶活性,降低β-AP,升高GSH-Px酶活性来实现对老年痴呆模型鼠预防和治疗作用。
Objective: To study the effect of Zhisheng Capsule I (ZSJN) on intelligence, monoamine oxidase (MAO), β-amyloid protein (β-AP) and glutathione peroxidase (GSH-Px) in Alzheimer’s disease model rats , to explore its prevention and treatment of Alzheimer’s effect and mechanism. METHODS: Rats with Alzheimer’s disease were made by intraperitoneal injection of D-galactose. Rats’ learning and memory abilities were determined by Morris water maze. The contents of MAO activity, β-AP and GSH-Px in brain were determined by biochemistry and radioimmunoassay. . RESULTS: Compared with normal rats, the learning and memory ability of the model group was significantly reduced. After the prevention and treatment of “Zhisheng Capsule I”, the learning and memory abilities of the model rats were protected and improved, the MAO activity in the brain tissue decreased, the content of β-AP decreased, and the GSH-Px enzyme increased. Conclusion: Zhisheng Capsule I can obviously enhance the learning and memory ability of model rats, and reduce the activity of monoamine oxidase, decrease β-AP and increase the activity of GSH-Px enzyme to achieve the preventive and therapeutic effects on Alzheimer’s disease model mice.
